- Intro to STORM 2 imaging with Colenso and Yari
- no nuclei on sample. 🙁
- Imaging en inv locus on Zeiss 700, tou-flu and en bio in yw 4 look pretty good. One good data stack taken for analysis.
- Test gel of DNA nicked translation gives no bands, despite 200-500 ng/uL spec reading. Too large / not digested? even extensive running doesn’t produce visible bands. Try PCR confirmation first.
- labels for second round embedding all lost in ethanol — should have written down color code, or write underneath label.
- samples difficult to extract in resin from complete-volume exchange tubes. pippetting works better than removing mesh. Should try with paint brush — need to buy brushes.
- got GPU code from Fang, need to try install and run on Tuck.
