- Etching 4H sample.
- Testing 4H sample on STORM.
- 4H has very weak switching. Might be new Glox not working. Or extra resin? see STORM post.
- Confocal of new in situs. en-Pc H3-750 failed. en still backgroundy in bio and still no luck with E(Pc) / tou-dig. I think these probes may have gone bad?
- en Psc has early germ-band elongation embryos with good engrailed and tou staining. H3-750 still undetectable by 405 — not a good sign for having the chromatin channel. Probably still a failure.
- Etch new 2H sample.
- Should try tiling at 4x.
- Added a few new 3D image processing scripts.
- Cytoplasm still fluoresces brightly under high laser power, though nuclei bleach quickly. Looks like resin still in tissue, even though it’s gone everywhere else. Better to etch longer, try 50 min next time. Problem on 50% of samples today.
