Laser powers
- 405: 1.5 mW
- 488: 115 mW at 45% power
- 561: 135 mW at 200 mW GUI/laserhead
- 642: 135 mW at 200 mW GUI/laserhead
- 750: 250mW at 1800 mA. –> too high?
- 750: 120 mW at 1400 mA –> too low?
- 750 190 mW at 1600 mA try this
Observations on switching
- 647 still switching rapidly with 4.8% 405 at 50K frames.
- 488 swtiching looks much better after ~40K frames. Still switching strong.
cryo-sections:
- cent-647 dramatically dimmer (due to overlong overhot hot washes? / lost probe?). 488 H3K9me2 very bright.
- sections very not flat
