Understanding bursty Gal4-LacZ expression

Researching UAS-LacZ line sequences to probe stochastic expression effects.

my UAS-LacZII from flybase stock 3955,

From Kathy Matthews, 1998 gift of flybase.

contains: P{UAS-NZ}20b — SV40 T NLS-lacZ fusion in pUAST (beta-gal is localized to the nucleus). Insertion on chromosome 2. From Yash Hiromi and S. West, unpublished.

LacZ on III BSC 3956,  is from same collection: P{UAS-NZ}J312 — Insertion on chromosome 3. From Yash Hiromi and S. West, unpublished.

comes from progenitor pUAST.  Map:

has 5x UAS-sites.

Reference: Brand and Perrimon 1993

Report comment: Contains 5 tandemly arrayed, optimized GAL4 binding sites (UAS\’s), followed by an Hsp70 TATA box and transcriptional start, a polylinker with 6 unique sites, and the SV40 small-t intron and polyA site.

Maybe it’s the TATA that makes the expression bursting instead.  Need to do a Drosophila promoter TATA box exploration.
