Sequential staining
- Attempting to align images from sequential staining. Correspondence not obvious.
- Substantial translation and rotation, need to fix these with feducials.
Probe making
- Run column cleanup on LC libraries after 10 round PCR using adapter primers (mismatched TM).
- moderate yield, eluted in 20 uL, ~20 ng/uL product. Should still be enough to seed a large scale PCR reaction.
STORM
- Finish O/N STORM run of en embryos
- Set up new O/N STORM run on en embryos
