Project 2
- Piranose: staining is still good in piranose treated cells. GA background is EXTREME, but certainly uniform, not punctate on the whole cell conventional level at least
Cell staining
- check cells from yesterday — all stains failed, including positive control
- too high hybridization temperature? (at least for the positive controls?)
- checked heat block temperature for new stain. Currently set at 85C is reading 65C — probably utterly insufficient for staining.
- Plate new cells
- Fixed, Triton treated, at 4C, ready for LN2 and Hcl.
- stained old cells not used yesterday + old embryos which failed BXC stain. Stained AATAT-P1, en-P1, and RNA-en-rcP1
Literature
- Reading new Corces/Taylor/Dekker lab’s 5C ES cell paper. Selected for journal club.
