- Finish O/N STORM of R2bxcB
- set up STORM of R2bxcD
- R2bxcD highly 405 responsive, low background, lots of switching (85K frames recording).
- Up’d laser power 200 mW, running near 100%. Need to measure again.
- Measured 40-45 mW at microscope backport appeture (at 60-80% AOTF, 200mW).
test RNA probes
- no obvious dots
- high cytoplasmic background, lower nuclear background (oposite of DNA probes)
- bright foci stuck around cells, around nuclei
- some potentional nuclear spots, quite weak
- LOTs of probe stuck in bright foci (clusters?) on the coverglass.
- very similar to my cell culture RNA in situs
- Try next embryo wholemount DNA FISH with RNA probes.
