Probe making
- test gel for samples 3 and 4 at 2:6 primer:RNA and 42 vs 50C incubation for 1hr.
- no incoroporation at all. Must have screwed something up.
Cell staining
- wash out stains for F11 double label and G1 double label test (using S3-Alexa750 this time).
- test stain on STORM2 — dots look good this time. 750A high background but mostly looks good
- Extra 20 min of 60C wash, see if this improves
STORM imaging
- need to max new Glox. (done)
- need to make new COT aliquots. Where did my pure COT go? (done, COT is in downstairs freezer)
- Imaging F11 sample. A750 visible but still pretty dim. Maybe try less BME next time?
