Chromatin Analysis log

• analyzed B04 up through image 20 (48 spots).
• analyzed B05 up through image 20 (30+ spots)

STORM imaging

• fix STORM acquisition of O/N imaging
• Finish STORM imaging of PHmut + ANTC + BXC
• Start STORM imaging of F01. Quite large for a 5 kb domain!

Probe making

• finished probe preps of L3 B10-12, C09-C12, and D01-D08.

PH project

Data

• copying data to Monet
• need to launch spot-fitting analysis

STORM

• imaging PHwt with BX-C and ANT-C after 10 min FA post-fix of anti-flag-647 label.
• qualitatively, 647 staining looks diminished relative to GA post-fix samples previously examined.
• hybes look great.

• Disk ran out of space, crashing B09 data acquistion after movie 30 (should still be plenty of data)
• cleared up disk space on Alistair Temp.
• copying B09 data from STORM2 to Monet
• imaging L3E11-F01 (piecewise reconstruction of L2G06)

Data Analysis

• calibrate parameters of B06 – B09 data
• Launch dot-fitting analysis (DaoSTORM3d single fitter) on B06-B08 on Cajal
• Launch fiducial fitting on (B06 and B07 data) on Tuck
