complete fluid exchange embryo rinse chambers

New method for rapid and complete fluid exchange during embryo washes
  • Cut a P1000 tip to fit snuggly inside a 1.5 mL eppendorf with the lid closed.
  • Cut a small whole in the bottom by removing an edge off the bottom with a razor blade.
  • squeeze a P200 tip into the bottom whole.  It should get pinched at the apperature, trim just above the pinch and you’ll have an insertable snug plug.  I’ve matched tip and tube with colored tape.
  • cut 1 inch square nylon embryo-sized mesh.  place ontop of cut eppendorf tube and insert cut P1000 tip.
  • Seal bottom of tube with parafilm before adding solution.
  • To rinse, remove parafilm with lid closed, apply modified P200 tip and remove liquid establish pressure, open lid to finish asperating dry.  Re-seal bottom before adding new solution.  Should not require letting embryos settle.

Developed similar version for resin embedding.  Need modified vaccum for collecting hazardous waste in separate tube.  (currently using 15 mL centrifuge tube punctured at top with a hyperdermic needle and with hose plugged into top of tube).
