- Sent snail bi-stability abstract to ML. Got feedback need to incorporated still
- Rehydrating embryos to test histone and pol-II antibodies last night: short RT primaries at 1:50 (polII) and 1:100 (H3K27 and H3K4). Secondaries polII m-488, and m-405:750. H3 is my new rab 647.
- Also running ON embryos at 1:250 polII and 1:500 H3 antibodies at 4C.
- Flies laying very well now, should be able to set up for some tighter collections.
- restriction digest minipreps with NotI (rare cutter?) or linearize with a different cutter? — can’t see wells without a backing — remember to add tape. Make sure gel is not deeper than comb — valley connecting wells not good for seperating DNA.
- double bands on all wells. didn’t run out an undigested control. Doesn’t look like NotI cut well, despite 3hr incubation. Or all fragments popped out similar size. Should have run control.
