- Finish O/N confocal run, all 9 embryos completed. Most are early cc14
- Analyzing O/N dot counting on old data with revised multilayer and peak-intensity checker. Basic counts all in the same range, some gradients a bit sharper (reduced background false-positive calls?)
- some orig images (MP10s from 2011-2-17) are somewhat underexposed for total mRNA, need to reduce peak intensity. Should choose dynamically from image.
- projecting data from last night — should flip order of tracks on 710 saved file to do 555 first.
- Accept review for PCB
- cut new cc14 H3K4me3 sample
- resin still not dissolving despite 20 min in sodium ethoxide and clear precipate upon second sodium ethoxide dip. Try different batch?
- set up new O/N run of MP10 het embryos (focusing on wt snail early dynamics still).
- more work on figure for dynamics analysis.
