STORM imaging 2/16/12
Laser calibration at output:
- 750 100 mW,
- 647 200 mW
- 561 100 mW (200 at laser)
- 488 200 mW
- 405 1.3 mW (max why low?)
- substantial resin still on slide S7 etched yesterday for 30 min.
- etched new samples from S1 and S7 (# 62 and 68 in records).
- first positions recorded on slide S1 lost when steve feezes. Retake 10x images.
- Identify germband elongated embryo captured previously in confocal images. Choose section from anterior extent ubx/AbdA expressing.
- Choose only 12 positions since run can only be 8 hrs.
- 20Hz 750, 60Hz 647, 30Hz 561 and 30Hz 488.
- shorter movies: 10,000 750; 27,000 647; 12,000 561; 10,000 488.
- time is calculated at 60Hz, so it will be twice: 7 hrs +1 for switching, should be done by 8A for Graham. Could have done more positions.
