- Staining with ch anti-GFP and rb anti-H3K27 O/N at 4C.
- STORM imaging on en, E(Pc), H3 and membranes (and tou if it comes out). ID 100+ en loci by 647-tagged E(Pc) expression. setup to image overnight. Conv images of dot channels, STORM of 750-H3
- Threshold for 750 is too low, weak background gives hazy objects rather than letting the switching events fill in properly.
- Transformed ligations of Dfd2, AbdB1, AbdB2, aib5E and + and – controls. 120 uL cells + 100 uL mixed IPTG + XGal (bottom of the aliquot). at 37 C O/N
