- Flip fly cages. Old Pc-GFP dying –> freeze.
- Need to characterize loss of snail mRNA in snail nulls better. ~early cc14 levels look like even half peak cc13 + degradation: ~85 transcripts per cell (highs just over 100). late cc14 20-30 transcripts transcripts per cell. Expect to drop to 10% (~10 transcripts/cell) in 30 min (close to background levels 5-15 range per nucleus, depending on stain quality). Measure embryos still in the 20-30 at cephalic furrow. Should account for a 10 – 20% transcription rate judging by presence of nascent transcripts. With 10% cells transcribing expect about 22 mRNA/cell after 30 min.
- working on z-color alignment. xy-alignment by nuclear lamina looks good. 750 and 647 beads getting mapped 1000 nm apart. Even when using the same calibration curve and by eye the distribution of x elongated and y elongated is equal. All mapped positions are also outside of range of sample. Maybe its the drift correction that’s miles off.
