  • Fixed z-offset issue: correct-to-beginning z-drift introduces wild errors.  Suggests that z-drift is not well tracked, will change drif correction parameters.  Without correcting z-drift at all alignment looks pretty good after warp.
  • association of 647-K27 and 750-SuZ in early germ-band extending embryos looks interesting.  Need control for just Dm01 double label.  SuZ12 present in several clusters, some of which contain K27 clusters some which lack.  Very few obvious K27 clusters lack SuZ12 at this point.
  • Flipped collection cages, 3:30P.
  • analyzing SuZ12-647 K27-750 data for comparison.
  • Need to develop script to identify and mask lamina boundaries.  These are very nice to have stained.  Width of laminin stain is 35-160 nm.  (nuclear pore 150, membrane ~50, backed by lamin layer ~50nm) See 3D SIM paper.
  • Pc-GFP data has similar density of Pc-750 and Pc-GFP 647 clusters in nuclei, substantially different than early embryo stains of K27 and SuZ for example.  None-the-less, substantially less overlap of clusters than expected, probably less than the K27 at SuZ loci frequency.
  • O/N embryos with SuZ Pc or SuZ H3 + hox genes are in 30% resin O/N.
