- Analyzing previous Pc-GFP images. z-calibration not as good as recent attempts with SuZ + K27.
- Stains to do: Pc-GFP stain Pc vs GFP in 647/750 colorflip with m-Dm01 647/750 background control in 5+ hr embryos.
- Discuss beam expansion with Colenso. distance between lens should equal sum of focal lengths. Beam expansion is equal to larger over the smaller focal length. Lens and mounts need to fit in tight gap between laser and mirror 1.
- Analyzing Antp data. control shows 1 strong K27 cluster next to 1 Antp dot localized. All other STORM videos lost focus :(.
- Analyzing Ubx AbdB data. Some suggestive clusters. High 647 non-nuclear background.
- Working on snail dynamics manuscript revisions.
- cross-correlation analysis of snail-endogenous and snail-yellow in wildtype background.
