- STORM run lost focus. Haven’t analyzed yet at which position / how many movies lost.
- Setup new collection of positions. All pretty old embryos in this batch, K27me3-647 stain doesn’t look great, channel has some background (probably resin etching issue, though some parts of the slide are overetched as well so not sure what to do here). Will definetely need a max consecutive frames to cut out some of the non-blinking points.
- Confocal scan of sna-488 in MP07 (minus prox) embryos still running smoothly by all appearances. Estimated completion around 10PM?
- lab meeting invitation from Francis group, discuss Petruk Cell paper (see post for notes).
- Confocal run finished and data saving, 9PM
- STORM run aborted 10:30 PM (illumination uneven, substantial non-bleaching features in most images) 🙁
