- Restart data run on Tuck
- Transfer files from STORM4 (en STORM from 9/6… oops).
- Etching slide S6 for imaging today.
- Move ND filter 5 from STORM2 750 line to STORM4.
- Colenso also notices switching issues with 647 channel but not others. Possible power density issue?
- STORM imaging of s6: E5 + AbdB (AbdB-488 not working well). 750 not switching as robustly as in previous samples.
- Running STORM analysis of AbdA data (8/25/12) and Pc data on Tuck.
- Yari wants full resources on Tuck for tomorrow, cancelled all Matlab ques to InsightM, hopefully most existing movies in process finish before tomorrow morning.
- Tuck update: 3 premature terminations (at frame 18,937 with 258,252 molecules found). Not a molecule number limit (I think that is 4 million, and some are still going fine at 480,000). 4 almost done (frame 100,000+ of 120,000). This is probably a sluggish switching problem.
- working on Dfd summary figures
- O/N STORM imaging of E5-Cy3B/AbdB-488 slide (s6). 647 providing good low background switching but switching slowly (Taking 60,000 frames, could probably due longer.
- Cajal also had about 10 movies crash InsightM. Probably system hit memory limit during acquisition which caused termination. Cajal is almost maxed out (CPU going at 60-70%). Just finished 750storm analysis hasn’t started 647storm (ran a few 647conv, shoud rewrite code to skip short dax files or things with conv in the name…).
