STORM
- characterizing embryo background (see images)
- 561 almost unsuable for STORM in embryos, lots of blinking and no respect for specific structures. Try at 1/4th power (edges of image look better in STORM). Excite embryonic background less.
- Background in 488 is extreme in STORM, pretty substantial in conventional
- 22 minutes, incomplete etching — sample bleaches down maybe 90% of brightness, but stops short of single molecule level. Attempting new coverslip at 32 minute etch.
- New coverslip looks okay started imaging. Set up O/N run.
