Research goals

Oligo Paints

  • Week of 01/15/13: synthesis en-Cy5 probe (by my hands in Zhuang lab and by Brian’s trainee at HMS) and of BX-C locus (by Brian).
  • Monday-Tuesday (01/21-22/13), test in situs of en and BX-locus
  • Tuesday: section Drosophila yw embryos to have samples to test en-OligoPaints in embryos.
  • Wed STORM en-locus (in S2 cells?) General Paint development on side

  • 01/16/13, start in situs trial 2 to test recently synthesized chr3R 500 kbp probe.

    • very weak signal detected on confocal.  Mostly cells too dense for good signal/good labeling. (cells near coverglass not accessable to probe because of biofilm on top).  

Coding

  • Add first last frame parameter choice to GUIDaoSTORM/GUIFitParameters for STORMfinder.
  • Integrate 3D bead calbration functions of STORManalyzer into STORMfinder, with more bead-displays
  • Integrate color warp map computation from STORManalyzer into STORMfinder.
  • ‘Light’ mode for STORMfinder (basically it’s current mode).  the z-fitting code and the color warpping codes have large kludgy libraries.  The codes are pretty fast (most of the time is all spent running InsightM thousands of times).
  • Shared clean version of STORMfinder with all default paths made globals specified by an external startup script.
  • Integrate merge conventional images function from STORManalyzer into STORMfinder.
  •  change sliders in Tiff viewer to have many more steps.
  •  check new code on Tuck with DaoSTORM
  •  basic viewing and running functions of STORMfinder are up and running
