STORM

  • O/N 200Hz run of BXC-cy5 still going
  • Not entirely sure this setting is actually running 200Hz.
  • dyes switching somewhat sluggish now, bumped up power to 300mW.
  • Stop run, image beads.
  • Prep new sample
  • 750 not switching well at all.
  • removed sample, will try again tomorrow night with TCEP?

Computer

  • shuffling data off of Buffalo Drive ‘Alistair3+3b’ failed during the night — drive disconnected. Might not have liked trying to transfer too many different ques over the same usb connection. Since that can’t actually be any faster to start with, might as well wait.
  • Reset transfers of Pho and Hb data to Alistair7.
  • Drives plugged into monitor get unmounted when monitor goes to sleep.

Embryo collection

  • Flipped all collection plates, 10:30 A
  • clean out of small collection plates need to make more
  • can’t find small petri dishes! – wrote to Alec about finding / ordering more. Maybe we ran out?
  • found petri dishes (back of cabinet). Made ~140 new small AJ plates
  • Fixed esc embryos and mat alpha tub x Psc1, Rad21, and Scm (O/Ns)
  • Fixed esc embryos and mat alpha tub x Pc3 (4-10hrs)

Fly work:

  • no virgin collection

STORM analysis

  • relaunch analysis of 02-02-2013 BXCemb data with new calibrations (from 02/14, original data was with 02/09 calibrations). I suspect more of these clusters are at the outer limits of the z-range than the current analysis suggests — some are clearly blurred in the conventional.
  • first pass analyzing embryo data (oops, applied to 488 — no wonder looks off)
  • applied automatic analysis to embryo data
  • annotated embryos (at least from dataset 2, where posterior/anterior is evident in some images).
