Check RNA hybes on STORM2

  • (hybed at 50C (55C?) in dry oven O/N in coverglass preps like for DNA labeling)
  • (quick check on turnkey looked promising)
  • tub looks good but hard to tell background vs. signal, hard to find nuclear dots. Substantially more dots though than Neu3.

Sectioning

  • Ultra-sectioning of Kc167 cells.
  • preferred cutting conditions: -50C knife/sample/chamber. 800 nm sections.
  • test sample looks promisingly like cut-sectioned nuclei on Turnkey, but cell density is surprisingly extremely low.

STORM

  • O/N STORM of en full-length cy5-secondary, 405 tertiary
  • much much brighter than previous stain
  • sealed with VALAP, imaged O/N.
