- 647 laser power seems lower — sample bleaching more slowly. Up’d to 180 mW, 80%-75% laser power.
- sample has moderate background and high to very high 405 responsiveness. Likely either cy5 primary + cy5 secondary or 405-primary cy5 secondary. Will see if other samples have more background.
- STORM analysis – can not run more than 11 versions of DaoSTORM from ProRAID on Cajal without major stall.
chromatin
- primer design finally finished clean run. ~1400 orthogonal 20mer primers with 14 or less bp homology to Human, Mouse, Rhesus Monkey, Drosophila or E coli, and 10 bp or less homology to others in the set. TMs all 70-80 range, no hairpins/secondary structure, have 3′ GC clamp (2 of last 5 bps must be GC).
