STORM Analysis
- E08 failed to run properly through split-dax
- launch STORM analysis. Running smooth at 70% on Cajal. Probably close to max read/write speed for disk drive across net. Wait to launch E08 again until tomorrow.
Cell Staining
- E10 y100 no heat denature
- D12 Y450 no heat denature
- RNase treat 4 coverslips (3 from 10/7, one from Bogdan’s samples last week) for 45 min at 37C. 10 uL of 100 mg/mL RNase A in 2 mL of PBS.
- RNase treat coverslips in prehybe solution (20 uL of 100 mg/mL RNase A in 10 mL)
- y100 10 ug RNase with heat denature
- y100 RNase no heat (double negative control for both RNase and non-denaturing)
Cell culture
- New cells growing slowly despite 28C (better than room temp close 18C.
- thaw 3 tubes of cells, combine together in single 5 mL
- letting cells recover at 28C.
- Concentrate on probe making this week, hopefully cells are kicking by the end of next week. Also try imaging multi-activator samples more.
