- Take GFP embryos out of 4C, incubating at RT
- heating up 2x SSCT for hot wash
- mounted oligo-paint embryos before and after washes (with WGA-488 for post-wash embryos). Remaining embryos at 4C
- Cut sections of Psc K4 at 330 nm. 3 slides worth.
- New water free sodium ethoxide seems to dissolve much better. samples look clear after 40 min.
- STORM imaging of en/tou/E(Pc) H3. H3-750 looks weak. some resin may affect swtiching in some embryos for 750? Greater challenge: finding the nuclear dots. Should make brighter probes (larger regions).
- 2 rounds of antibody labeling for GFP embryos. Washing out antibody, embryos at 4C O/N.
- Resin embedding 12 samples (139-150, see records). embryos in 70C 0vernight (8:10P)
