- Finish reviewing manuscript, write up review for submission. Still need final read-over and submit.
- Revising snail manuscript, working through results section.
- Gel extracting probe templates with KE.
- Confocal imaging, test stains. Pre-wash oligo-stained embryos have weak nuclear background with dense foci (under strong 647 excitation). Repeat at higher concentration without hot-wash steps. Try shorter hybe.
- Test GFP stains. en looks good. eve looks good. Pc looks Excellent
- miniprep frozen pellets. Bad prep – little DNA. Scrapped. Restart from transformation with frozen ligate.
