Fly Work

  • flip collection cage, 9:50 A
  • Collect virgin Mat-alpha-tubs, esc/CyO x da[10]/Gla and esc/esc mutants.
  • two new bottles of mat-alpha-tubs cleared and added to virgin collection
  • 1 hatched larvae in Mat-alpha-tub x SuZ(12) :(. large number of eggs, will see if these hatch. Flipped into new vial kept at 29C. (these F1s started to get crossed at RT, so not all eggs in first vial at 29C are from heat-activated mothers.
  • PM collection of mat-alpha tubs.
  • Check mat-alpha-tub-VP16 crosses: flipped out parentals.

STORM Analysis

  • Running 02-08-13 bead data through analysis. Good, pretty tight z-calibration curves. Z-range was strongly skewed to the + side for 750 and 647 data though. see post on chromatic warp for curves and for DaoSTORM fits vs. insight.
  • setting up parameters for 02-08-13 data.
  • Comparing chromatic calibration from DaoSTORM and Insight.

Coding

  • Reworking STORMrender
  • embedded figure axis for main plot, navigation plot, and chromatic histograms (could hijack this for other histograms as well).
  • switched handling of multicolor 3D to use cell arrays (much easier).

STORM

  • set up imaging of fab7-647 and ss-488 to run over night. calibration image looked pretty good.
  • running with new batch of glox and hard TIRF (14.4). contrast of 647 looks better than on last weeks images. Maybe glox was expiring? or maybe the better TIRF angle (instead of 14.2 and 14.3 range).
  • On run 3, 488, cells not visible. dropped to 14.33 TIRF angle, cells now visible with good switching. Hopefully the quality of the cy5 switching doesn’t drop to much.
