STORM
- Finish O/N confocal
- More cells not in TIRF – sections not bleaching, even 647. Dropped TIRF angle to 14.2, now switching well, still good contrast.
- Might need to re-image this slide later today.
- This data set will require manual curation of STORM movies (probably a good idea for all data sets, shouldn’t take too long with STORMfinder).

STORM Analysis
- running analysis of bead data from Fab7-647, Pc-750 run
- Cajal auto-rebooted last night (Go Windows! Always trying to ruin things).
- Fix: prevent Windows 7 automatic reboot: run gpedit.msc (Computer Configuration > Administrative Templates > Windows Components > Windows Update). Enable the setting that reads “No auto-restart with logged on users for scheduled automatic updates installations”.
- Relaunch analysis on Cajal in Insight.
- DaoAnalysis delayed.
- Found issue in z-calibration, preventing good calibration fits for the -600 to -200 range of motion (first movement of stage). Fixed and updated.
- Ran chromatic calibration for BXC embryo data.
Coding

Critical Data issue
- Alistair 8 stopped responding. EMERGENCY DISK MANAGEMENT NEEDED!
- temporary power loss (powersupply disconnected), disk no longer recognizes connection. Contacted HUIT.
- after powering drive off using the power button and restarting the disk, it now connects by USB. We’ll go back to trying to get the eSATA to talk to this disk tomorrow.

STORM
- imaging BX-C 2x Cy5, flanking region between ss, in 488. BX-C looks strong 120K frames easily (with some 405 activation), flank looks bright but not switching as much (more like fab7 level switching, 20-30K frames).
