Probe making
- Designed primaries and secondaries for oligoprobes, see:
SecondarySeqDesign_131001.m - Ordered 3′ Amine modified (add Cy7) and Cy5 modified secondaries and 5′ Cy3 modified primaries from IDT
- ordered A405 modified L2P3 primary from Invitrogen
- Setup for Deep sequencing of library: Scheduling qPCR training (MX3000)
- alliquoted primer dilutions (1:20) of T7 library into new plate (rpt PCR at 20 uL scale for whole library to test with deep-seq)
STORM
- Make new Glox buffer
- Set up imaging of G10 (Black 425kb chrX)
- staining looks good.
- switching is very good, 245,000 frames of good switching.
