General STORM stuff

Exploring effect of height and bead size on z-calibration curves

  • sonicating some 25 nm 580 beads, diluted 1:5000 in Ca2+ Mg2+ PBS buffer.
  • STORM2 very difficult for this — a little z drift and the crazy slow z-stage makes taking calibration very difficult
  • Took several movies scanning at different depths
  • Took several manual movies
  • 1:5000 is way too concentrated for the 25nm beads.
